ANALYSIS OF INTRACELLULAR PURINE METABOLITES BY HPLC IN DEVELOPING RAT CALVARIA

Abstract
The present experiment was performed to investigate the contents of metabolites involved in the purine catabolic pathways in developing rat calvaria. The portions of calvarias containing frontal and parietal bones was separated from 19--dayold rat foetuses. Specimens were assayed for their contents of intracellular purine metabolites by high performance liquid chromatography in the acid extract of the tissues with rapid quenching. The content of inosine was the highest in the purine nucleosides and bases. The content of adenosine was higher than that of guanosine, but xanthosine was not detected in the tissues. The contents of hypoxanthine and guanine were relatively high, but adenine and xanthine were not detected. These results implied that the major purine metabolites in developing rat calvaria were inosine, hypoxanthine and guanine.
